Summary of Expertise:

Listed with other top experts in:

Expert has spent most of his career conducting research on the chemistry of the synthesis of peptides. He has experience with all the common methods of coupling, which includes carbodiimides, mixed anhydrides, and phosphonium salt-based reagents, and he has contributed to the elucidation of the mechanisms involved. He is responsible for establishing the existence of 2-alkoxy-5(4H)-oxazolones as intermediates in coupling reactions. He has served as consultant to a process development laboratory. He is writing a book for learners on peptide synthesis that includes the latest developments.

Expert has devoted much of his research contributing to our understanding of the stereomutation that accompanies coupling reactions. He is the recognized authority in the field. He can provide the best advice as to how to minimize or eliminate the problem. He is responsible for introducing the more appropriate term enantiomerization instead of racemization, which is gradually being adopted by the peptide community.

He has experience with the preparation and use of derivatives of nearly all the amino acids and has published research papers on the development of many of these methods. He serves as consultant to a retail producer/supplier of amino acid derivatives.

Expert developed the modern chemistry of the difficult to handle N-methylamino acids. In 1973 he devised the simple method that is the current one for preparing synthetically useful derivatives of N-methylamino acids from the unmethylated derivative. He eliminated the misconception that N-methylamino acids are more chirally stable than unmethylated amino acids and established that it is the opposite. He has published a dozen research papers on mono-, di-, and trimethylation and has written review chapters on the subject.

Expert has spent all of his career in the field of amino-acid and peptide chemistry. He is well acquainted with derivatization and reactions of amino groups and the preparation of methyl esters. He has extensive practical experience with reactions involving activation and derivatization of carboxylic acids, in particular the preparation of esters, hydrazides and amides of amino acids. This involved amongst other fundamental contributions on the chemistry of mixed anhydrides.

Expert has experience in the separation, detection, and quantitation of enantiomers of amino acids, their derivatives and peptides, and the preparation of the single-isomer compounds. He has developed a method for converting a small peptide into the single-isomer form of its amide and methylamide via an active ester generated from a mixed anhydride. He has published numerous papers on the chemistry involved in the isomerization of amino acids and peptides and is an expert on the options available for minimizing it during the manipulation of these compounds. He has published numerous papers focusing on the changes in stereochemistry and their prevention that occur during the reactions involved in linking amino acid derivatives to form peptides.
