Expert has a broad background in biochemistry, having been involved in teaching various aspects of biochemistry to PhD candidates and medical students for over 40 years. Throughout this time he has always conducted a research program in which biochemical knowledge and techniques were applied to problems in oncology, hematology, burn and traumatic injury, and immunology. His studies have involved protein purification (enzymes and antibodies), enzyme assays, immunochemistry, radiochemical techniques, tissue culture, chromatography, spectrophotometry, electrophoresis, and others.

Much of Expert's research has been cancer oriented. His laboratory was instrumental in bringing asparaginase into clinical use for treatment of childhood acute leukemia. His group demonstrated that the catalytic activity was necessary but insufficient for therapeutic utility because certain physical chemical properties were also required. He also demonstrated the biochemical basis for resistance to asparaginase, the immunosuppressive activity of the enzyme, and development of immunity to leukemia after certain mice were cured of their leukemia by asparaginase therapy. The latter observation led to extensive studies of tumor immunity with cancer immunotherapy as the objective. A clinical immunotherapy trial was instituted in patients with metastatic renal cell carcinoma with autologous vaccines prepared in Expert's laboratory.


Expert's immunologic studies focused on two areas: malignant cells and human erythrocytes (RBC) as part of his experience with blood banking. Malignant cells were maintained by serial transplantation in syngeneic mice or by tissue culture, as were human cells. These cells were used for vaccine production, the study of adjuvants to enhance immune responsiveness, and as targets for antibodies. Antibody detection for these and other studies was by immunofluorescence, cytolysis, ELISA, radial diffusion, precipitation in multi-well arrangements, and others. Using blood typing antisera, studies with RBC were by agglutination in saline, colloidal media, with anti-globulin reagents, or enzyme altered RBC; complement fixation has also been used. The mechanism by which treatment of RBC with certain enzymes permits agglutination with IgG anti-Rh antibodies was elucidated.




Blood has been the focus of much of Expert's research. His career in biomedical science began in association with a blood bank where he gained knowledge of blood banking techniques, determined blood types, and used blood plasma as raw material in developing techniques of protein purification by ion exchange and molecular sieve chromatography. He has performed enzyme analyses on RBC and WBC, measured lymphocyte blast transformation in response to mitogens, and used radiolabled cells (including platelets). Most recently he has cooperated in an experimental cardiopulmonary bypass program exploring techniques for improving the compatibility of the oxygenator and accompanying circuits for blood. The success of this endeavor was measured using radiolabled WBC and platelets.

More recently Expert developed an interest in certain problems involving biomaterials. The function of endothelial cells was characterized on synthetic materials used as vascular prostheses to replace clogged blood vessels. His studies included the determination of cell binding to the prosthesis, cell proliferation, production of factors involved in blood coagulation, and susceptibility to attack by neutrophils. The potential of nitric oxide to inhibit platelet and neutrophil activation by synthetic surfaces in vitro and in vivo in a cardiopulmonary bypass model has been elucidated. He demonstrated the enhancement of nitric oxide effects by phosphodiesterase inhibitors. The latter finding shows how combining biochemical factors may be used to produce a synergistic effect.

Expert has taught a course entitled "Science Ethics and Societal Concerns" at Southern Methodist University, and has a number of other classes that relate to the area of bioethics. He takles the issues of insurability and employability based on genetic makeup. He sees the potential polemics than will occur if employers don't become informed in this area. Expert also consults at the personal level on the issues of euthanasia, family planning and abortion. In addition, he is knowledgeable of the safety of genetically engineered products, patents on genes and gene products, cloning, stem cell research, handling of (human) biological materials and exposure and handling of potential carcinogens.
