Summary of Expertise:

Listed with other top experts in:

Expert is a Bioscientist with expertise in molecular biology, protein biochemistry, antibody engineering, and enzymology. Extensive experience in wheat embryo cell-free protein expression technology. Outstanding communication, writing, and presentation skills. He is a tenacious problem solver and performs well in multi-disciplinary drug discovery and R&D laboratories. He is analytical and articulate.

During his doctoral studies, he has successfully stabilized many commercially valuable proteins through understanding of their thermal behavior. He also constructed biosensors for the estmination of pesticides in the environmental samples.

For his Post-doctoral studies he joined to the world renown Lab, where the primary focus of lab was to develop a wheat germ cell-free protein expression technology and commercialize it. Now this product is available for researchers world-wide. In this lab Expert focused on the constrcution of engineered antibodies and express them in the cell-free environment. Since post-translational modification like disulphide formation in the antibody demands oxidative conditions, he has adopted an innovative protocol to express functionally active single chain antibodies against pathogens and successfully characterize them. He is also well versed with antibody optimization techniques like ribosome display and synthetic library construction.

Upon moving to the United States, Expert established wheat germ cell-free protein expression technology starting from the scrap within a span of six months. He used his established technology to address the Malaria recombinant protein expression issues. Expert expressed more than 25 recombinant proteins with a 100% success rate. During these studies, he has used many techniques for the analsis of recombiant proteins, Such as, genetic engineering, Cell-free RNA expression, Dialysis, protein purification, assay development, SDS-PAGE and more.

Acting Instructor

Perform multiple functions, including assay development, inhibitor screening and targets substrate, and specificity testing of newly-expressed malaria drug targets. Established-assays were based on radio-chemicals, gels, thin layer chromatography, spectrophotometer, and more. Construct new genes from parasite RNA, cloning, and wheat germ cell-free expression of targets. • Expressed malaria targets when conventional expression systems failed at rate of 85%. • Directed key project with more than 90% authority. Trained graduate and undergraduate students. • Expedited expression of 25+ malaria drug targets with functional characterization of 100%. • Attracted collaborators/clients to lab and enhanced reputation of lab in research community. Data was well-received in various conferences, including ASTMH. • Improved protein expression yield, producing 20 synthetic genes with 100% success. Achieved by determining cause of protein fragmentation problem and recommending codon optimization by means of synthetic gene to avoid protein fragmentation.

Visiting Scientist

Instituted powerful and emerging cell-free protein expression technology based on wheat embryos. Achieved via generation of protocols to make high-quality lysate from scrap (e.g., wheat). Published articles and presented data at scientific conferences. • Planned and led project concerning cell-free protein expression technology. Trained undergraduate student to identify and characterize novel druggable site on validated drug target. Work was important due to combination of established cell-free technology and inhibitor screening. • Delivered entire process of high-quality wheat germ lysate preparation and formulation with various buffers, salts and amino acids for optimal protein expression within 4 months with investment of less than $5,000, saving thousands of dollars in wheat embryo lysate costs. • Confirmed power of cell-free system by expressing collaborators' proteins, which were not possible to express in cell-based expression systems. • Confirmed role of calcium in protein by means of biochemical experimentation that was not possible even with crystal structure

Cell-free Sciences and Technology Research Center

Researcher

Advanced development of wheat germ cell-free technology over 1-year period and over 2 subsequent years, adopted wheat germ cell-free protein expression technology to produce disulphide bonded engineered antibodies. Constructed mouse and human antibody libraries against malaria parasite. Developed ribosome display method to select for high affinity antibodies against desired target. Trained medical graduate student from Thailand. • Collaboratively published 2 book chapters on applications of cell-free protein expression technology. • Expressed functionally-active antibody fragments by formulating oxidative conditions to create disulphide bonds. Further system was established to produce large quantities of these antibody fragments successfully. • Contributed to construction of antibody libraries. • Devised innovative protocol to formulate oxidative conditions to produce disulphide bonded proteins in cell-free environment without compromising yield of expressed proteins. Protocol led to production of many malaria vaccine candidate proteins.
